The ThermoFisher Glacios 200 kV Cryo-TEM System at the ULS is equipped with 200 kV high-brightness field electron gun (X-FEG), an autoloader, and as well as a ThermoFisher Falcon 4i direct electron detector (DED). By taking advantage of the latest generation of DED, the Glacios delivers higher image quality and shorter total image acquisition time than its predecessors. The system can be used for both single particle analysis (SPA) and cryo-electron tomography (cryo-ET). With improved hardware and a new enclosure that minimizes environmental influences, the Glacios can deliver near-atomic resolution with an information limit of up to 2.1 Å. In addition, transmission loss over 24 hours due to ice growth has been reduced to 2%, allowing researchers to collect more high-resolution images with less interruption. On the other hand, the Smart EPU Software enables faster and easier setup and processing, benefiting both novel and experienced users.

Sample screening is an essential component of successful cryo-EM workflows, allowing verification of sample quality in terms of both biochemistry and vitrification. The Glacios fits seamlessly into single-particle analysis and cryo-ET workflows with convenient and contamination-free sample transfer. The Glacios at the ULS will be mainly used for cryogenic sample screening and cryo-ET data collection.
