The Illumina MiSeq system offers a straightforward, user-friendly solution for next-generation sequencing (NGS). It supports small scale projects or longer sequencing read length. Different from the classic Sanger chain-termination method, MiSeq applies 4-channel sequencing by synthesis technology (SBS) to perform the sequencing. It allows the fluorescent-labeled single bases incorporating into the growing DNA strands, and each of the four DNA bases was identified with different fluorescent dyes (A with red, T with green, C with yellow, and G with blue). Fluorescent dyes are imaged when each base is added to the growing DNA strands. Base calling can then be proceeded from the fluorescent signals at each cycle.
The MiSeq works with 4 different sequencing kits (Nano v2, Micro V2, Standard v2, and v3), which meets different application requirements. By applying the Standard v3 sequencing kit, MiSeq allows the maximum output of 25M of reads and 15 Gb for each run.
